Hetzel, FW (Health One, Colorado, USA) Abstract PDT requires O2 within the target tissue during laser irradiation. We have shown that tumor response rate could be improved by combining hyper-oxygenation and PDT. This study is to investigate the effect of oxygen delivery route, i.e., vasculature or superficial diffusion, in the combined treatment modality. Implanted mammary carcinoma bearing legs of C3H mice were tied with rubber(s) band to cut off the local blood flow in various treatment groups. The results show a clear decline in tumor response rate when local tumor blood flow is cut off during PDT treatment, compared to those from the control groups which received normal PDT treatment. The tumor response rate did not diminish for the PDT/rubber band treatment group when the procedure was performed in combination with hyper-oxygenation. This indicates that superficial diffusion may play an important rule in PDT treatment combined with hyper-oxygenation.
