(2nd Dept.of Oral & Maxillofacial Surgery, Tokushima University School of Dentistry, Japan) Abstract TS-2 monoclonal antibody which neutralizes interferon(IFN)-g-inducing activity of OK-432, a streptococcal preparation, was generated. TS-2 also recognized lipoteichoic acid (LTA) derived from Streptococcus pyogenes. We purified the LTA-related molecule (OK-PSA) which induce IFN-g on human peripheral blood mononuclear cells (PBMC) from the butanol extract of OK-432 by an affinity chromatography on CNBr-activated sepharose 4B-bound TS-2. The stimulation of PBMC with OK-PSA induced IFN-g, interleukin (IL)-12, tumor necrosis factor (TNF)-a, TNF-b and IL-1b in a dose dependent manner. The cytolytic activities against cancer cells were also accelerated by the treatment with OK-PSA. Although natural killer cells produced IFN-g in the absence of monocytes as well as in the presence of them, the secretion of IFN-g by CD4+ and CD8+T cells was limited in the presence of monocytes. Only few amounts of IL-10 were induced by OK-PSA, and it was shown that the major component of OK-432 for IL-10 production was streptococcal M-protein. These results clearly indicated that OK-PSA, LTA-related molecule isolated from OK-432, was potent inducer of Th1-type cytokines.
