Abstract

SERCA pumps sequester Ca2+ into the sarco/endoplasmic reticulum and from there it is released during signal transduction. The uptake occurs with a stoichiometry of 2 Ca2+ per ATP hydrolyzed. SERCA proteins are encoded by three genes (SERCA1, 2 and 3) located on separate chromosomes. SERCA transcripts are expressed and alternatively spliced in a tissue dependent manner. The resulting mRNA species encode different SERCA protein isoforms and differ in 3'-untranslated regions (UTR). SERCA protein isoforms differ in their Ca2+ affinity, resistance to oxidative stress and modulation by sarcolipin, phospholamban, and Ca2+/calmodulin kinase II . This poster will summarize this information and then analyze it by posing the following questions. How does Ca2+ affinity of the pump affect tissue function? Do some tissues need more oxidation resistant SERCA isoforms than others? What is the importance of a phospholamban binding site in SERCA1 and 2 but not in 3? Do the isoforms differ in their localization with respect to Ca2+ release channels, agonist induced Ca2+-entry and/or Ca2+ binding proteins? Does the SERCA pump isoform expression relate to properties of Ca2+ wave forms produced upon cell activation? - Supported by Medical Research of Canada and Heart & Stroke Foundation of Ontario.
