Administration of alpha-Lipoic Acid Overcomes Detrimental Effects of n-3 PUFAs on Wound Healing in Rat

Abstract

As previously shown dietary n-3 polyunsaturated fatty acids suppress the connective tissue proliferation in the rat liver wound with high level of lipid peroxidation. In the present study influence of alpha-lipoic acid as a natural antioxidant on the n-3 fatty acid induced effects has been investigated. Animals were given commercial rat pellet diet (control group) or diets enriched with 10% of sunflower oil (n-6 group) or 10% of cod liver oil (n-3 group) for 8 weeks followed by addition of alpha-lipoic acid to the same diets at the calculation of 30 mg per rat for 10 days before the liver thermic wound was induced. Lipoic acid administration was continued after wound infliction for 6 days, then in the liver wound the proliferation of the connective tissue, the level of lipid peroxidation and peroxidizability, the content of prostaglandins (PG) E2 and F2alpha and the redox status was estimated. Lipoic acid prevented n-3 fatty acid induced suppression of the connective tissue proliferation in the healing wound, avoided increase of peroxidation of lipids, reduced peroxidizability of lipids, modulated the decrease of PGE2 and PGF2alpha and improved the redox status. The results indicate that dietary alpha-lipoic acid may prevent n-3 polyunsaturated fatty acid induced adverse effects.
