Age-Related Decrease in BDNF in the Zitter Rat Brain

Abstract

The zitter rat exhibits several neurological abnormalities such as body tremors and flaccid paresis in the hind limbs. Neuropathologically, morphological alteration of the zitter rat is characterized by sever vacuolation (edematous spongiform encephalopathy) and hypomyelinatation in the central nervous system (CNS), and progresses with age. On the other hand, brain-derived neurotrophic factor (BDNF) is a member of the neurotrophin family, and support the survival of a specific set of neurons. Moreover the BDNF has been noted that this neurotrophin holds therapeutic potential for treating various types of injury to the CNS. To investigate whether BDNF is associate with these pathologies in the zitter rat, we analyzed BDNF mRNA level in zitter rat brain compared with Wistar-tremor control (WTC) rat. BDNF mRNA from rat brain aged 3,10,30,80 days were examined by Nothern blotting and Slot blotting. Expression of BDNF gene in the zitter rat brain was decreased to half of that in age-matched WTC rat. This result is suggested BDNF may be associated with neuronal degeneration in zitter rat brain.
