Invited Symposium: Quinones and Other Reactive Oxygen Species in Neurobiologic, Apoptotic, and Neurotoxic Processes |
Abstract Cultured rat fibroblasts,murine neuroblastoma and monkey kidney tumor cells(Vero line) were exposed to dopamine or dopaminochrome in the presence and absence of ascorbic acid.The exposure was made for different times up to 2 hrs and different concentrations up to 200 micromolar of dopamine or dopaminochrome.The levels of DT-diaphorase and glutathione were also measured in the cells, together with those of dopamine peroxidase,a new enzyme activity found in this laboratory in rat brain extracts.It appears that the bioreductive defences of the cells are correlated with their ability to withstand oxidative injury. Ascorbic acid seems to act as a pro-oxidant in the presence of dopaminochrome thus potentiating its toxic effect.
