In your opinion, is in situ fixation at physiological pressure a reliable method to evaluate changes in vascular tone, lets say in response to acute infusion of a vasoconstrictive agent such as an alpha-adrenergic agonist?
Your study suggest that fixation itself may introduce an artefactual reduction in lumen size. Can this be controled ?
