Abstract Increased levels of endopeptidases such as cathepsin D have been demonstrated to be useful in the prognosis of breast cancer (BC). However, little attention has been devoted to other classes of proteolytic enzymes such as aminopeptidases (AP). To evaluate the clinical role of AP in BC, we measured six AP activities in serum from women with BC, and compared these values with a control group. In addition, serum 17-beta-estradiol and prolactin levels were measured in BC patients. Serum AlaAP, ArgAP and GluAP were significantly lower in BC patients than in controls. There exists a significant inverse correlation between 17-beta-estradiol levels and AlaAP, ArgAP and CysAP. These results suggest that aminopeptidase activities are estrogen-inhibited and contrast with other estrogen-induced proteases used as BC markers which are overexpressed in BC. The simultaneous presence of hypersecreted proteases with decreased levels of AP may be useful in the diagnosis and prognosis of BC.
