Electrochemotherapy Potentiation of Antitumor Effect of Cyclophosphamide by Local Electric Pulses on the Metastatic Lesion of Hamster Oral Fibrosarcoma

Abstract

Electrochemotherapy is a new antitumor treatment, in which electrical pulses are given to the tumor site several minutes after injection of antitumor chemical agents. In this study, oral fibrosarcoma was submucosally transplanted into the cheek pouch mucosa in the hamster. After transplantation, the metastatic lesions appeared in the regional lymph nodes of hamsters. When the tumor size in the metastatic lesion was approximately 100 mm3, treatments were initiated. The animals with metastatic lesions received an intraperitoneal injection of cyclophosphamide(CPA), antitumor agent, followed by local delivery of electric pulses to the metastatic site using BTX electroporation unit. The tumor was markedly reduced in size several days after this treatment. CPA injection alone or electric pulse treatment alone had no significant suppressive effect on the tumor growth. These results clearly indicated that the antitumor effect of CPA on the metastatic lesion of hamster oral fibrosarcoma could be considerably potentiated or enhanced by local electric pulses.
