Dr. Koylu: Great presentation. Hope you have fun at the meeting. I have some difficulty in understanding what you are measuring in these experiments. The rats received an NO inhibitor- this inhibitor is not going to produce a downregulation of NO synthase but just a change in activity. Does the inhibitor remain in the isolated membranes afterwards during your glu uptake experiments? I am not clear on why you would expect a permanent change in the glu uptake and not just a change as long as NO is present if NO is to act as a messenger. However, if you think that NO produces an irreversible damage, then the blockage of normal NO synthase activity should not have any effects anyway. I am not clear.
