I am confused. Can you explain how iNOS induction due to treatment with L-NAME can cause 5-HT hyperreactivity? You have stated that 5-HT hyperreactivity can be explained by the deficiency of NO to counteract the 5-HT contractions. Your experiments with L-arginine suggests that L-NAME induces iNOS, therefore there should be an increased local amount of NO.
